چکیده

A symbiont, isolated from root nodules of Crotalaria saltiana Andr., was identified as Rhizobium sp. It produced large amount of extracellular polysaccharides (EPS) in broth culture in yeast extract mannitol (YEM) medium. The EPS production and growth started simultaneously, though they have different stationary phases. The symbiont produced maximum EPS (16 μg/mL) when the medium was supplemented with preferable carbon (glucose) 2%, nitrogen (glycine) 0.1%, and vitamin (biotin) 1 μg/mL. Attempts were made to optimize the cultural requirements for growth and EPS production. The EPS produced by the symbiont contained glucose and galactose. The possible role on Rhizobium root nodule symboiosis has been discussed in the light of EPS production.

Production of Indole Acetic Acid by 5-fluro-tryptophan Resistant Mutant of Rhizobium Sp. Isolated from Root Nodule of Pulse Legume Phaseolus Mungo L

A symbiotic strain was isolated from the root nodules of the legume pulse Phaseolus mungo L and designated as PM 25. The strain was identified as Rhizobium sp. on the basis of the physiobiochemical characteristics and 16S rDNA sequence based homology. The Minimum Inhibitory Concentration of the analogue 5-fluro-tryptophan was 3.0 mM. The analogue (5FT) resistant mutants were screened from wild ...

International Journal of Pharma and Bio Sciences PROTEIN PROFILES OF IN VITRO AND IN VIVO HALOTOLERANT RHIZOBIUM SPECIES ISOLATED FROM CAJANUS CAJAN PLANT

Salinity-induced changes in the protein profiles in Rhizobium sp. exhibited alterations in proteins, which either showed an enhanced rate of synthesis or a decline in the levels as compared with controls. The in vitro and in vivo Rhizobium of Cajanus Cajan plant produced high amount of protein from halophillic glyphosate (Hg) as compared to the Normal (N) plant. The proteomic diversity contribu...

Screening of Alginate Lyase-Producing Bacteria and Optimization of Media Compositions for Extracellular Alginate Lyase Production

Background: Alginate is a linear polysaccharide consisting of guluronate (polyG) and mannuronate (polyM) subunits. Methods: In the initial screening of alginate-degrading bacteria from soil, 10 isolates were able to grow on minimal medium containing alginate. The optimization of cell growth and alginate lyase (algL) production was carried out by the addition of 0.8% alginate and 0.2-0.3 M NaCl ...

International Journal of Pharma and Bio Sciences GROWTH BEHAVIOUR AND INDOLE ACETIC ACID (IAA) PRODUCTION BY A RHIZOBIUM SP. ISOLATED FROM CAJANUS CAJAN PLANT

The Root nodules of Cajanus Cajan plant produced high amount of (35.56 ug/gm) of Indole acetic acid from Hg plants as compared to the N plant. The IAA was measured maximum from Rhizobium spp. of C.cajan plant at 24h, when the bacteria reached the stationary phase of growth. Mannitol & KNO3 preferred to be a good carbon and nitrogen source for IAA production in all N, Ng, H & Hg Rhizobium strain...

The exopolysaccharide of Rhizobium sp. YAS34 is not necessary for biofilm formation on Arabidopsis thaliana and Brassica napus roots but contributes to root colonization

Microbial exopolysaccharides (EPSs) play key roles in plant-microbe interactions, such as biofilm formation on plant roots and legume nodulation by rhizobia. Here, we focused on the function of an EPS produced by Rhizobium sp. YAS34 in the colonization and biofilm formation on non-legume plant roots (Arabidopsis thaliana and Brassica napus). Using random transposon mutagenesis, we isolated an E...
